Guanidine hydrochloride protein denaturation

Guanidine hydrochloride (GndHCl) is a powerful chaotropic agent. It not only promotes the solubility of hydrophobic molecules, but also denatures proteins. In a high-concentration (6 M) guanidine hydrochloride solution, the ordered "secondary structure" (maintained by intermolecular non-covalent interactions such as hydrogen bonds and hydrophobic bonds) of almost all proteins is destroyed, and the peptide chain is stretched. The tertiary structure was changed.

At room temperature, the maximum solubility of guanidine hydrochloride is 6 M, and the appearance is a colorless solution. If 8 M solution needs to be prepared, heat treatment at 35 ºC is required for about 30 min.

Usage

Guanidine hydrochloride is mainly used as an intermediate in medicine, pesticides, dyes and other organic synthesis. Guanidine hydrochloride is an important raw material for the manufacture of sulfadiazine, sulfamethazine, sulfamethazine and folic acid. It can also be used as an antistatic agent for synthetic fibers. In addition, guanidine hydrochloride can also be used as a detection reagent for detecting precious metals such as gold, iridium, osmium, platinum, and ruthenium.

Guanidine hydrochloride protein denaturation protocol

There are two degeneration mechanisms of guanidine hydrochloride. One is the solubilization of guanidine hydrochloride on hydrophobic amino acid residues. The other is that the denatured protein can preferentially combine with guanidine hydrochloride to form a denatured protein-denaturant complex. When the complex is removed, the reaction equilibrium moves to the right, and the protein in its natural state continuously transforms into a complex, which eventually leads to complete denaturation of the protein. However, the combination of denaturant and denatured protein is very weak, and only a high concentration (6M) of denaturant can cause complete denaturation of the protein. In general, the denaturation caused by guanidine hydrochloride is usually reversible.

Guanidine hydrochloride RNA extraction
